In this experiment, the effect of temperature on reaction rate of enzyme will be studied. Peroxidase will be used as test enzyme and the variable to be studied is temperature (°C). At first, the experiment will be carried out at room temperature of 20°C. The temperature of the experimental setup will then be increased by keeping the reactants in a water bath at a temperature of 50°C. The enzyme catalysed reaction rate will also be studied at a lower temperature by placing the reactants in an ice bath to bring down the temperature to 0°C.

The measuring cylinder was filled with water and another shallow dish was also filled with water and kept ready.
After this, the measuring cylinder was placed underwater in the dish and turned upside down. Following this, the delivery tube was inserted from the bottom of the measuring cylinder.
The level of water in the measuring cylinder was noted. The meniscus reading was carefully taken to avoid parallax error. A delivery tube and cork was inserted into the flask and then sealed.
Peroxidase enzyme solution was taken using a 1mL syringe. A stopwatch was kept handy to note the time.
As soon as peroxidase was added to the substrate, i.e. hydrogen peroxide, in the beaker, the stopwatch was started.
The enzyme and substrate solutions were mixed gently and the volume of oxygen resulting from the reaction was noted after 20 seconds.
The above step was repeated by placing the flask into high temperature water bath as well as ice bath. The level of oxygen was noted both at 50°C and 0°C. Care was taken to keep the enzyme concentration, substrate concentration and pH constant.
