Define magnification and resolution.
Magnification is the degree to which the size of an image is larger than the object itself. The number of times bigger the image is than the object.
Resolution is the ability to distinguish between to objects that are very close together. The higher the revolution the greater the detail.
How do light microscopes work and what is their limitations on magnification and resolution.
Use a number of lenses to produce am image. Light passes under the stage through the condenser lense. Usually have x4,x10,x40 and x100. The eye piece magnifys a futher x10.
Capable of x1500. Maximum resolution is 200nm.
What is staining and sectioning?
Coloured stains are chemicals that bind to chemicals on or in the specimen so it can be seen. Acetic orcein stains DNA dark red. Hentian violet stains bacterial cell walls.
Spcimens are embedded in wax, thin sections are then cut withou distorting the structure of the specimen. Useful for soft tissue.
