Androgens
Testosterone
Testosterone esters
17-alpha-alkylated androgens
Drugs to block Androgen Activity
Receptor Inhibitors
5-alpha reductase inhibitor
Gonadotropin-related drugs
GnRH agonists
GnRH antagonists
Testosterone
Testosterone esters
17-alpha-alkylated androgens
Uses: Hypogonadism of hypothalamic/pituitary origin and gonadal failure or insufficiency
Dosage: begin slowly and gradually increase; establish lifetime maintenance
Problems: can not be given orally (except 17-alpha-alkylated androgens, but HEPATOTOXIC)
- better given transdermally
Receptor Inhibitors (Flutamide)
5-α Reductase inhibitor (Finasteride)
Helps block formation of androgen that supports prostate growth and hyperplasia (BPH)
GnRH agonist (leuprolide)
GnRH antagonist
Chemical castration
Agonist - given chronically will desensitizes receptors so won't produce more hormones
Given with androgen receptor antagonist to prevent disease flare at initiation of agonist, and blocks effects of adrenal androgens
