Brompheniramine, chlorpheniramine, azatadine, cyproheptadine, clemastine, diphenhydramine, premethazine, meclizine- Class, MOA, Pk, AE, Use
Class: 1st generation H1 antagonist
MOA: reversible and competitive inhibition of H1 receptors
PK: oral, 30 min onset, liver metabolism
AE: SEDATION, anticholinergic, N/V, increased appetite, and wt gain, cardiac toxicity
Use: can use for motion sickness and sedation
Acrivastine, azelasatine, cetirizine, desloratidine, fexofenadine, and laratidine- Class, MOA, PK, AE, Use
Class: 2nd gen H1 antagonist
MOA: comp and rev inhibition of H1 receptor
AE: not as many as 1st gen
Use: rhinitis, conjunctivitis
Cimetidine, Famotidine, Ranitidine, Nizatidine- MOA, AE, Drug Intxn, Use, Which had worst SE profile?
MOA: rev and comp inhibition of H2 receptors-- parietal cells especially
AE: libido, gynecomastia, confusion
Drug Intxn- CIMETIDINE inhibits p450- phenytoin, theophyllin, phenobarb, carbamazepine, CCB, quinidine, warfarin
Use: ulcers, ZE syndrome, GERD
Pseudoephedrine, phenylephrine, oxymetazoline, naphazoline- MOA, PK, AE, Use
MOA: direct alpha agonist, vasoconstriction
PK: quick onset w/4-6 hr duration, partial hepatic metabolism, excreted unchanged by kidney
AE: oral-- cns stim, HTN, arrhythmias, URINARY retention, BPH, topical--nasal irritation, and rebound congestion
USE: oral not as effective but longer duration, less local AE, topical has lots of rebound congestion-- can only use for 3 days
