Mucus Layer Microbiome

Improved Essays
The mucus layer microbiome is a part of the fish immune system, it is the first line of defense in protecting the fish from harmful external pathogens. The mucosal layer of the fish microbiome is examined to gain insight into the structure of the fish skin mucosa. In this experiment, an unknown strain of bacteria was isolated from the mucus layer of a Gambusia affinis, and then thoroughly tested to identify the FBI. The mucosa microbiome of the Gambius affinis is home to thousands of bacteria, however, by means of serial dilutions, the solution was made less concentrated to the point where a single cultured sample could be able to isolate one single bacteria. Then FBI 926 was isolated and further microbial identification test was used to paint

Gram Negative Unknown Lab Report #20 BIOL 3444-008 Giang Nguyen TA: Malini Kotak 11/05/2014 Abstract The experiment is conducted in order to identify a randomly given unknown gram-negative microorganism. By comparative analysis of biochemical tests that were performed throughout the semester to find out its characteristics and determine the identification of bacterium out of four potential unknown was given. To figure out the characteristics, T-streak was performed to isolate the sample from broth to a Tryptic Soy Agar plate, along with an additional slant in case something wrong with the original plate that can possibly affect the result.…

Discovering Unknown 41 The overall reasoning for this experiment is to figure out what the unknown bacteria is through a process of tests and research. There are several differential media and selective & differential medias used throughout my findings, and all were done using aseptic technique. Based on the results of my test I came to a conclusion that my unknown 41 is Kocuria rosea.…

The next test, Bacitracin Susceptibility, was the next test on key that helped lead to the identification of the unknown. For this procedure, you grab a Blood Agar plate, and then streak the plate with a flamed loop containing your unknown. After the plate is streaked, you will then ask your instructor to place antibacterial discs throughout the plate. These antibacterial discs, after growth, will show the zone of inhibition which will then lead to your results. You leave the plates for a couple days in order to later on see some growth.…

The study of microbiology requires not only understanding the microscopic organisms, but also the understanding of lab techniques and procedures used to identify, control, and manipulate microorganisms. The identification of microorganisms is not only important in microbiology lab, but also in the medical field to identify an agent of a disease that will help treat the patient by using the correct antibiotics to kill off the host. In this unknown lab report, techniques and procedures learned in the microbiology laboratory during the semester that was performed to test ones practical understanding of microbiology. The sole purpose of the unknown lab is to demonstrate understanding of the experimental methods and lab techniques learned during…

Information about the Bacteria Pseudomonas aeruginosa During microbiology lab, an unknown bacteria culture in liquid broth was assigned to be identified by conducting a series of various tests. Nearly twenty different tests were conducted on the bacteria, but the most important of these was Gram staining test, gelatin stab test, and oxidase test. The results of these three tests allowed for the determination of the bacteria genus and species.…

To determine the unknown’s morphology is the first step in narrowing down possible organisms. A MacConkey and Mannitol Salt Agar tests were conducted on the organism and revealed the following. No growth on the MacConkey plate resulting in a gram positive bacteria and the Mannitol Salt Agar result showed it could be Staphylococcus or Micrococcus. (1) A gram stain test was used in determining the shape, size and arrangement of the unknown bacteria (1). The unknown organism is heat fixed onto the slide, in order for the organism to be stained.…

The lab instructor issued out a test tube labeled with letter ‘I’, which consisted of two unknown bacteria, Gram-positive or Gram-negative that were streaked from a pure culture. Sterile techniques were followed while performing precise instructions as stated in the referenced Laboratory Manual. Example 1: The first procedure performed was done by isolating a pure culture from the mixture onto a solid Trypticase soy agar (TSA) media. Sterile technique was done by flaming the loop until it turned red to ensure that there were no current bacteria on the loop avoiding contamination followed by rapidly flaming the neck of the test tube to prevent the entry and contamination of unwanted microbes.…

In order to determine the identity of two unknown bacteria, I performed a series of experiments over several days. To begin this process I created a dichotomous key that I utilized as a guide to perform a series of inoculations, while using both selective, and differential media. The series of inoculations eliminated bacterium based on test results until the correct bacteria were both positively identified. On day one of this process I received two bacteria, one labeled 7A and the other labeled 7B. I began the experiment with thirteen possible bacteria.…

The properties of selective and differential media suggest that differences between the bacterial species could be used in this lab to identify these six species but their effectiveness is unknown. The efficacy of identifying a completely unidentified bacterium in a cost-sensitive manner using these test also has yet to be determined. We hypothesized that applying a combination of selective and differential media with biochemical tests would provide a presumptive identification of the first group of six unknown bacteria. We also hypothesized the identification an unknown member of the Enterobacteriaceae family referred to as unknown #53 could be definitively identified using the API 20E and other tests. To test the first hypothesis, samples of four of the six unknown bacteria were streaked onto plates of Mannitol Salt agar, MacConkey agar, and Eosin Methylene Blue agar to examine their growth on the media.…
