It denatures the bacteria so it sticks better to the slide.
Is the dye Methylene Blue acidic or basic?
Basic (positively charged).

The bacteria is negatively charged so a basic dye will attract better to it.
Why is the negative staining procedure better than the simple stain when observing morphologies?
The negative stain does not stain the bacteria, but only the surrounding area. Dyeing the bacteria could alter the morphology.
What dye was used in the simple stain?
Methylene Blue.
What dye was used in the negative stain?
Nigrosen.
