Is the acid-fast stain medically important? If so, how important in relation to gram staining?
Yes and in relation to the gram stain, it is second in importance.
Why is acid-fast staining important?
Acid-fast staining is important because of the highly pathogenic nature of certain members of the genus: Mycobacterium such as M. leprae and M. tuberculosis.
Why does the genus "Mycobacterium" not completely destain during a gram stain process?
Because of a high concentration of mycolic acid in their cell wall which produces an irregular effect on the second example of a gram-positive organism.
Does the mycolic acid in the genus "Mycobacterium" make it easier for an acid-fast stain to identify?
Yes.
What does an acid-fast positive cell look like?
Red.
Why do acid-fast positive (also known as acid-fast) cells retain their red color?
Because the carbolfusin in an acid-fast positive cell is more soluble in the cell wall lipids than in acid-alcohol.
What do acid-fast negative cells look like?
Blue.
Why do acid-fast negative (also known as non-acid-fast) cells appear blue?
Because acid-fast negative cells lack the lipid components and the carbolfushin is rapidly removed during decolorization which leaves the cell colorless. The acid-fast negative cell appears blue after methylene blue is applied as a counterstain.
What is used as a mordant for an acid-fast stain and what cell layer does the primary stain (carbolfuchsin) penetrate?
Heat and the carbolfuchsin penetrates the waxy mycolic acid layer.
